摘要
目的 观察冈田酸(0A)对SH—SY5Y细胞tau蛋白磷酸化的影响。方法 MTT比色法观察OA对SH—SY5Y细胞代谢率的影响,免疫细胞化学法和蛋白免疫印迹法观察OA对SH—SY5Y细胞磷酸化tau蛋白水平的影响。结果 40nmol/LOA孵育SH—SY5Y细胞24h可显著降低细胞代谢率,其效应随着OA浓度的增加和孵育时间的延长而增强。OA作用于细胞3~48h,蛋白免疫印迹显示Ser-396位点磷酸化tau蛋白水平逐步升高,24h达高峰,之后下降。免疫细胞化学法显示,OA作用于细胞24h,Ser-396位点磷酸化tau蛋白水平较对照组细胞增强。结论 OA增强SH—SY5Y细胞tau蛋白Ser-396位点磷酸化水平。
Objective To oberserve the effect of okadaie aeid(OA) on phosphorylated tau in SHSY5Y cells. Methods MTT method was used to measure cells viability after OA treatment. Western Blotting and immunocytochemical staining were used to detect the level of phosphorylated tau in SH-SY5Y cells. Results Treatment of SH-SY5Y cells with 40 nmol/L OA for 24 h caused a decreased in the metabolic rate of cell [(63.8±2. 6) % vs control 100% ,P〈0. 05]. 40 nmol/L okadaic acid induced tau hyperphosphorylation in SH-SY5Y cells. The levels of phosphorylated tau at site Ser-396 was increased after exposure of 3, 6, 12 h and reached the maximum level after exposure of 24 h. Conclusion OA can induce tau hyperphosphorylation in SH-SY5Y cells, which ascribed to the pathogenesis of Alzheimer's disease.
