摘要
利用杂交瘤技术制备人胰腺γ-GT酶(PGGT)单克隆抗体,并采用间接混合夹心法(ELISA),以PGGT单克隆抗体检测临床病例血清271例,正常人血清对照40例,鉴别诊断胰头癌、肝癌、胆管癌和胆管良性梗阻。结果发现:PGGT单克隆抗体诊断胰头癌的敏感性、特异性分别为100%和99.6%;正确率99.6%;阳性预测值、阴性预测值分别为92.3%和100%,优于PGGT/TGGT,CA_(50)和CEA的诊断方法。因此,PGGT单克隆抗体检测PGGT是特异性诊断胰头癌更为理想的免疫学方法。
PGGT monoclonal antibodies were prepared by hybridization technique. The sera of 217 patients and 40 normal controls were examined by using ELISA with PGGT as antibodies for differentiation diagnosis of carcinoma of head of pancreas, liver cancer, malignant tumors and benign obstruction. The results revealed that the sensitivity, specificity and precision of PGGT method in diagnosis of cancer of head of pancreas were, respectively, 100% and 99.6% with precision being 99.6%. The negative ancl positive prediction values from the PGGT method were 92.3%and 100%, which were better than those obtained by PGGT/TGGT, CA_(50) and CEA techniques. We believe that PGGT MAb technique is a better immunological method for the specific diagnosis of cancer of head of pancreas.
