对前列腺癌的基因放射联合疗法的生物学效应观察

摘要 目的:探讨基因治疗和放射治疗联合应用于前列腺癌的治疗效果和安全性。方法:应用单纯自杀基因,射线照射及联合方法分别处理2种前列腺癌细胞,alamarBlue还原法观察细胞生长情况,绘制细胞生长曲线。结果:由细胞生长曲线可以看出较低浓度的基因转染和低剂量的照射联合应用,获得了比常规剂量的单纯治疗更好的治疗效果。结论:联合治疗中两种治疗可以相互促进,低剂量的联合应用就可以达到甚至超过常规剂量的单纯治疗,同时剂量的降低也提高了治疗的安全性,为进一步研究和临床应用提供了实验基础。 Objectives:To evaluate the efficacy and safety of gene therapy combined with radiotherapy for the treatment of prostate cancer.Methods:two prostate cell lines were treated using suicide gene,radiation exposure alone,or combined therapy of suicide gene and radiotherapy.Cell growth was determined by AlamarBlue reduction assay,and a growth curve was drawn.Results:The cell growth curve suggested that a combination of low-concentration gene therapy and low-dose radiotherapy was more effective in inhibiting the growth of prostate cancer cells than the normal dose of either gene therapy or radiotherapy alone.Conclusion:Gene therapy and radiotherapy show synergistic action in their combined treatment for prostate cancer.Low-dose combination might obtain the same or even better efficacy of either therapy of normal dosage.
