摘要
目的观察临床常用的4种补益方剂对N-甲基-N-亚硝脲(MNU)诱导的光感受器细胞凋亡的影响。方法SD大鼠分为正常组、模型对照组、四物汤组、杞菊地黄汤组、四君子汤组和肾气丸组。TUNEL法检测光感受器细胞的凋亡,常规HE染色测量视网膜外核层的厚度。结果与模型对照组相比,四君子汤和肾气丸组大鼠在细胞凋亡和外核层厚度方面无差异,而四物汤和杞菊地黄汤组大鼠光感受器细胞凋亡百分率有不同程度下降,同时外核层厚度显著增厚。结论四物汤和杞菊地黄汤能抑制MNU诱导的光感受器细胞凋亡,且杞菊地黄汤疗效优于四物汤。
OBJECTIVE To study the influence of four nourishing Chinese herbal recipes for treatment of N - methyl- N- nitrosourea (MNU) - induced apoptosis of photoreceptor cells. METHODS Sprague Dawley (SD) rats were divided into six groups: the control group, the model group, the sijunzi (四君子) decoction group, the siwu (四物) decoction group, the qijudihuang (杞菊地黄) decoction group, and the shenqi (肾气) pill group.The photoreceptor cells apoptosis was marked with TUNEL. The thickness of outer nuclear layer (ONL) was measured by HE coloration. RESULTS Compared with the model control group, the apoptotic indexes and the thickness of ONL of the sijunzi decoction group and thes shenqi pill group had no significant difference, whereas that of the siwu decoction group and the qijudihuang decoction group had significant difference. CONCLUSIONS The siwu decoction and qijudihuang decoction could restrain MNU - induced apoptosis of photoreceptor cells. The therapeutic effect of qijudihuang decoction was better than that of siwu decoction.
