摘要
用含NaH2PO4浓度分别为0、0.2、0.4、0.6、0.8、1.0、1.2 mmol/L的营养液处理后,平邑甜茶根系质膜H+-ATPase的活性随着根区磷浓度的升高而升高,并在1.0 mmol/L处达到峰值;该酶的对硝基苯磷酸(PNPP)水解活性也明显升高,其Km值由3.48 mmol/L下降至0.97 mmol/L;同时结合抑制剂试验,钒酸钠对幼苗根系质膜H+-ATPase的抑制效应也得到了加强。
After treated with the nutrient solution containing 0,0.2,0.4,0. 6,0.8, 1.0, 1.2 mmol/L NaH2PO4 respectively, the activity of root plasma membrane H^+ -ATPase in Malus hupehensis (Pamp) Rehd. was obviously enhanced and reached the peak value at 1.0 mmol/L. PNPP hydrolysis activity was also promoted by phosphor,and its Km value decreased from 3.48 to 0. 97 mmol/L. And it was also found that 1.0 mmol/L NaH2PO4 could raise the inhibition of H^+-ATPase activity by sodium vanadate. These results indicated that phosphor might regulate the activity of H^+- ATPase by means of affecting the phosphatase domain of this enzyme.
