摘要
报道了用高效液相色谱法电化学检测器测定羟基游离基的方法。用ODS(3μm)柱,0.03mol/L的醋酸钠和0.03mol/L的柠檬酸钾(pH3.80)缓冲液为流动相,以水杨酸为捕获剂,微透析采样,测定了前列腺素造成大白鼠脊髓损伤时羟基游离基的释放。
Hydroxyl free radical(OH) reacts with salicylic acid(SA) and generats 2, 3- and 2, 5-dihydroxybenzoic acid (DHBA), which can be measured electrochemically in picogram quantity by HPLC. The column used was Phase Sep Ltd. ODS (3μm) 150mm×4. 6mm and mobile phase was 0. 03mol/L solium acetate and 0.03mol/L potassium citrate (pH 3. 80). Male Spraque-Dawley rats(300-400g) were used. When rat was fully anesthetized, its back was shaved and laminectomy was performed from vertebra T12 to vertebra L3. The sciatic nerve was exposed. Except for 2mm dialysis zone, microdialysis fiber was pull through the cord. A solution of 5mmol/L SA in ACSF was pumped and after 75min the collection of control sample started. Fluid was collected continuously from outlet of the fiber. Alter collecting two 30-min control samples, a solution of 2mmol/L PGF2a.and 5mmol/L SA in ACSF was administered into the rat spinal cord through a microdialysis fiber. The products of OH attack on SA, 2,3- and 2, 5-DHBA from microdialysate increased by 3. 6 and 2. 6 fold respectively(n=5). Combining these results with our previous findings we know that OH generated by administration of Fenton reagent into the spinal cord caused PGF2α. release. It is demonstrated that there is a feedback mechanism between OH and PGF2α. Increase of one induces the increase of the other. In MP-treated group, 30mg/g body weight of MP was given through the toil artery at the time when control samples were collected. The results demonstrated that MP completely blocked OH generation.
