摘要 目的观察四逆散有效部位对嗅球损毁大鼠海马细胞发生的作用及CREB基因拷贝数量的影响。方法以细胞分裂标记物胸腺嘧啶脱氧核苷类似物Brdu腹腔注射,采用免疫组织化学方法显示和计数海马Brdu标记阳性细胞;使用MGB探针,采用TaqManTM技术,定量检测CREB基因拷贝数量。结果四逆散有效部位明显增加了海马齿状回(包括SGZ和hillus区)Brdu标记数量,同时增加了CREB基因拷贝数量。结论四逆散有效部位治疗通过增加CREB基因表达,对抗和逆转应激对海马神经元发生的抑制作用,从而发挥抗抑郁作用。 OBJECTIVE To investigate the effect of Sini powdered effective composition on hippocampal neurogenesis in the rats following the lesions of olfactory bulb. METHODS The Brdu positive cells were marked by using the thymidine analog bromodeoxyuridine ( Br- dU), a maker for cataloging cells, then showed and counted by immunohistochemistry. CREB gene was quantitively tested on the basis of Taqman^TM technology using MGB probe. RESULTS Chronic Sini powdered effective composition treatment significantly increased the number of BrdU-labeled cells in the dentate gyms and hilus of the hippocampus and CREB copy number. CONCLUSION Sini powdered effective composition treatment as antidepressant could oppose or reverse the action of stress on the proliferation of hippocampal neurons by increasing the expression of CREB gene.
