乌司他丁对重症胰腺炎大鼠T淋巴细胞凋亡的影响 被引量:6

The Effect of Ulinastatin on Apoptosis of T Lymphocytes in Rats Induced by Severe Acute Pancreatitis

摘要 目的:探讨乌司他丁对重症胰腺炎大鼠T淋巴细胞亚群及各亚群凋亡比例变化的影响。方法:Wistar大鼠32只被随机分成假手术对照组、重症胰腺炎(SAP)模型组、乌司他丁治疗组和地塞米松治疗组。利用向胆胰管内注射脱氧胆酸钠建立大鼠重症胰腺炎模型。采用直接免疫荧光法先对CD4+和CD8+T淋巴细胞进行标记,使用原位缺口末端标记法标记各T淋巴细胞亚群的凋亡细胞。通过流式细胞仪检测实验结果。结果:乌司他丁治疗组CD4+T淋巴细胞比例和CD4+/CD8+比值较SAP模型组明显回升(P<0.05或P<0.01),CD4+和CD8+T淋巴细胞凋亡比例均明显下降(均P<0.01)。结论:乌司他丁可能通过某种途径减少T淋巴细胞的凋亡从而发挥减轻重症胰腺炎诱发免疫抑制的作用。 Objective: To investigate the effects of ulinastatin on the apoptosis of T lymphocytes in rats induced by severe acute pancreatitis. Methods: Thirty-two Wistar rats were randomly divided into the sham-operated control group, the severe acute pancreatitis (SAP) group, the ulinastatin group and the dexamethason (DXM) group. The severe acute pancreatitis model was established by using intrapancreatobiliary duct injection of sodium deoxycholate. Percentages of CD4^+, CD8^+ T lymphocytes were labeled by using direct immunofluorescence assays, while the apoptotic ratio of acinar cells were determined by using methods of insitu end labeling(TUNEL).At last flow cytometry was performed to investigate the result. Results: The percentage of CD4^+ T lymphocytes and the ratio of CD4^+/CD8^+ in the ulinastatin group were significantly higher than that in the SAP group (P〈0.05 or P〈0.01), the ratio of apoptotic CD4^+ and CD8^+ T lymphocytes were all significantly decreased (P〈0.01). Conclusion: Ulinastatin may relieve the immunosuppression in SAP by decreasing the apoptosis T lymphocyte.
