普罗布考和氯沙坦对兔血管成形术后细胞增殖的影响

Effects of probucol and losartan on cell proliferation after balloon angioplasty in rabbits

摘要 目的研究普罗布考和氯沙坦对兔血管成形术后细胞增殖的影响。方法雄性新西兰白兔40只,随机分为高胆固醇组、普罗布考组、氯沙坦组和联合用药组。行髂动脉球囊拉伤术,用免疫组化检测血管胰岛素样生长因子-I受体(IGF-IR)内皮生长因子(VEGF)和增殖细胞核抗原(PCNA)表达。结果与高胆固醇组相比,普罗布考组、氯沙坦组和联合用药组管腔面积明显扩大(P<0.01),内膜面积明显减少(P<0.05),IGF-IR、VEGF和PCNA表达明显减少(P<0.01),但三组间差异无统计学意义(P>0.05)。结论普罗布考和氯沙坦可以通过抑制IGF-IR、VEGF和PCNA的表达显著减轻兔髂动脉血管成形术后再狭窄的形成。而联合用药与单独用药之间无差异。 Objective To investigate the effects of probucol and losartan on cell proliferation after balloon angioplasty in hypercholesterolaemic rabbits. Methods Forty male New Zealand rabbits were randomly divided into high cholesterol diet group, probucol group, losartan group and combined drugs group. The iliac arteries of the rabbits were balloon - injured and the insulin-like growth factor-I receptor ( IGF-IR), vascular endothelial growth factor ( VEGF), and proliferating cell nuclear antigen (PCNA) were detected by using immunohistochemical method. Results Compared with high cholesterol diet group, the lumen areas were enlarged and the intimal areas were decreased in the probucol group, losartan group and combined drugs group (all P 〈 0. 05). The expressions of IGF-IR, VEGF and PCNA were significantly decreased in the probucol group, losartan group and combined drugs group than those of the high cholesterol diet group ( P 〈 0. 05). Conclusion Probucol and losartan can prevent the restenosis of rabbitg iliac artery after balloon injury through inhibiting the expression of IGF-IR, VEGF and PCNA. There is no significant difference on the curative effects between combined drugs and single drug administration.
