干扰素-β对胶质瘤的生长抑制以及对其他细胞因子的调节作用的初步研究 被引量:2

摘要 目的研究干扰素-β(IFN-β)对胶质瘤细胞系SK-MG-1的生长抑制作用和诱导凋亡的作用。并研究IFN-β在SK-MG-1细胞系中对其他细胞因子的调节作用。方法分别用IFN-β基因和蛋白刺激胶质瘤细胞系SK-MG-1,通过MTT方法检测细胞增殖情况,并运用流式细胞技术检测其对凋亡的影响,同时用实时定量PCR的方法检测细胞因子的变化。结果结果显示IFN-β可以抑制细胞增殖,诱发细胞凋亡,定量PCR结果显示在胶质瘤细胞细中IFN-β上调白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的mRNA表达,同时一过性下调白细胞介素-6(IL-6)的mRNA表达。结论IFN-β可以抑制胶质瘤细胞系SK-MG-1的增殖,诱发细胞的凋亡。可以调节其他细胞因子的表达。 [Objective] To study the growth regulation effect and cytokine regulation effect of interferon -β(IFN-β) on ghoma cell line for the purpose of further studies on IFN-β gene therapy on gtioma. [Methods] We stimulated the SK-MG-1 cells with IFN-β protein and IFN-β gene transfection with liposome. After that we used MTT method and flow cytometry to investigate the growth inhibition effect and apoptosis induction effect of IFN-β on SK- MG-1 cells. Then real-time PCR method was used to detect the cytokine network induced by IFN-β. [Results] IFN-β protein and gene could suppress the growth of SK-MG-1 cells and induce apoptosis. We also found that interleukin-6 (IL-6) expression reduce transiently after IFN-β stimulation but interleukin-1β(IL-β) and tumor necrosis factor-α (TNF-α) expression were up regulated. [Conclusion] IFN-β can suppress SK-MG-1 cell growth and induce apoptosis. It can also activate cytokine network in this cell line.
