摘要 目的研究醛固酮(aldosteroen,ALD)及其受体拮抗剂螺内酯(spironolactone,SPI)对大鼠肾小球系膜细胞转化生长因子β1(TGF-β1)基因表达的影响。方法以不同浓度的ALD(10-11、10-9、10-7mol/L)及/或10-7mol/LSPI刺激系膜细胞24h后,应用半定量RT-PCR方法检测细胞TGF-β1mRNA的表达;以10-9mol/L浓度的ALD刺激系膜细胞不同时间(12h、24h、48h)后,应用半定量RT-PCR方法检测细胞TGF-β1mRNA的表达。结果半定量RT-PCR结果显示,ALD促进培养的大鼠肾小球系膜细胞TGF-β1mRNA的表达,且具有浓度依赖性和时间依赖性的特点,SPI能够拮抗ALD的作用。结论ALD促进肾小球系膜细胞TGF-β1的基因表达,进而导致肾小球硬化。SPI能够拮抗ALD的作用,从而可能有益于延缓肾小球硬化的进展。 [Objective] To evaluate the effect of aldosterone (ALD) and spironolactone (SPI, an aldosterone receptor antagonist) on the expression of TGF-β1 mRNA in cultured rat mesangial cells. [Methods] Mesangial cells were treated with medium containing different concentrations of ALD (10^-11, 10^-9 and 10^-7 mol/L) and/or 10^-7 mol/L SPI for 24 h, while control cells were treated with vehicle only. The expressions of TGF-β1 mRNA were detected by semi-quantitative RT-PCR. Mesangial cells were treated with medium containing 10^-9mol/L ALD for different time (12 h, 24 h and 48 h). The expressions of TGF-β1 mRNA were detected by semi-quantitative RT-PCR. [Result] The expressions of TGF-β1 mRNA were increased significantly by ALl) in a dose- and time-dependent manner. SPI inhibited the stimulative effect of ALD. [Conclusion] ALD up-regulates the expression of TGF-β1 mRNA in cultured rat mesangial cells. Aldosterone antagonist inhibits the effect of ALD, and thus implicates in the treatment of Glomerulosclerosis.
