摘要
实验选用4个地方品种:清远麻鸡、惠阳三黄胡须鸡、封开杏花鸡、广西霞烟鸡以及3个近年培育的品系:岭南黄快大Ⅱ号配套系、矮小鸡E4专门化品系、AA配套系为材料(每个品种或品系各取5个个体,共35个个体)。采用PCR一鲫结合DNA测序技术。对H—FABP(心脏型脂肪酸结合蛋白)基因第2外显子区进行了单核苷酸多态性(SNP)分析。结果表明:在第一内含子区含有丰富的SNP、位点:580bp(T→G)、595bp(A→C)、610bp(C→-)、617bp(A→G)、622bp(A→T)、625bp(A→-)、627bp(A→C)、629bp(A→C)、631bp(A→C)、677bp(G→A)和1个双碱基突变586-587bp(TG→CA);在第二外显子区发现了一个非SNP位点785bp(C+T);这表明在外显子2上本实验所选品种的序列之间没有差异,但与GenBank上登录号为AY648562的序列相比存在变异。此外在第二内含子区也发现了一个非SNP位点:1018bp(C+T)。这为进一步分析H—FABP基因的遗传变异及其与内质性状的相关性以及将该分子标记应用于育种计划奠定一定的理论基础。
In this study, the single nucleotide polymorphisms (SNPs) in extron 2 of H- FABP gene in 35 individuals from 7 different chicken breeds (strains) Qingyuan Ma chicken, Huiyang Sanhuang Huxu chicken, Fengkai Xinghua chicken, Lingnanhuang Fast-growing Ⅱ chicken, Dwarf E4 chicken, Guangxi Xiayan chicken and Arbor Acres boiler were detected by PCR- SSCP methods and sequence analysis technique. The results show that there are abundant SNP sites distributed in intron 1. These SNP sites are located at 580 bp (T→G), 595 bp (A→C), 610 bp (C→- ), 617 bp (A→G), 622 bp (A→T), 625 bp (A→-), 627 bp (A→C), 629 hp (A→C), 631 bp (A→C), 677 bp (G→A), and 586-587 bp (TG→CA) is also found; In extron 2, a non - SNP site is found which demonstrates that there was no variance in DNA sequence of extron 2 in different breeds (strains) tested in this study, but genetic mutation exists in extron 2 area compared with the template sequence. In addition, a SNP site is also found in intron 2 area at 1018 bp (C→T). All of these findings are good theoretical foundation for the progressive research of the relationship between genetic mutation in H- FABP gene and meat trait and for the progressive research of the application of molecular markers in animal breeding.
