化瘀生脉方对内皮细胞缺血缺氧保护作用的实验研究

摘要 为研究化瘀生脉方对体外培养的内皮细胞缺血缺氧损伤模型的保护作用。将化瘀生脉中药兔血清培养液、正常兔血清培养液分别加入缺血缺氧损伤的内皮细胞培养,分别称为化瘀生脉组、空白对照组。另外,将正常兔血清培养液加入正常内皮细胞培养,称为正常组。进行细胞死亡率的检测,细胞培养液中乳酸脱氢酶的测定,并于透射电镜下观察内皮细胞的形态结构变化。结果显示,3组细胞死亡率、LDH漏出量方差分析均为P<0.001;正常组内皮细胞死亡率、LDH漏出量最小,空白对照组最高,化瘀生脉组低于空白对照组。3组均数两两比较,P<0.01,差异均有显著性。透射电镜下见,化瘀生脉组较空白对照组粗面内质网数目较多,无扩张,其上附着的核糖体存在,平滑内质网扩张程度减弱。表明化瘀生脉方对内皮细胞有一定的保护作用。 The objective of the paper is to study the protective effect of stasis-eliminating and pulse-activating(SEPA) recipe on ischemia-and anoxia-caused injury(IACI) of endothelial cells(EC) in culture in vitro.Endothelial cells with IACI were divided into the treated and the blank control groups,which were cultured in the medium containing normal rabbit serum and SEPA TCD and in the medium containing normal rabbit serum alone,respectively,and the normal endothelial cells were taken as the normal group and cultured in the medium containing normal rabbit serum;the cell death rates and lactic dehydrogenase(LDH) leakages of all groups were measured and the morphological changes of CE observed under electronic transmission microscope.The results showed that the variance analysis of the death rates and LDH leakages of the three groups showed P < 0.001: the normal group had the least death rate and leakage,the blank control had the highest and the treated had the ones lower than those of the blank control;the two-to-two comparison of the three-group means showed a significant difference (P < 0.01); it was found under electronic transmission microscope that in the treated group,the rough surfaced endoplasmic reticulum increased in comparison with that in the blank control and had the attached ribosome but no expansion;the smooth endoplasmic reticulum expansion decreased,suggesting that SEPA recipe has a somewhat protective effect on IACI of EC.【
