摘要
目的观察腰痛舒胶囊对骨关节炎模型中p53、bcl-2、增殖细胞核抗原(PCNA)、转移生长因子-β1(TGF-β1)的影响。方法采用免疫组化染色,在光镜下观察p53、bcl-2、PCNA、TGF-β1的表达。结果正常组大多数标本未见p53与bcl-2的表达,造模后在软骨移行层见到散在的p53与bcl-2的表达,用腰痛舒后bcl-2的表达有逐渐增强、p53的表达有逐渐减弱的趋势;正常组有少量PCNA阳性细胞的表达,造模组PCNA在滑膜表达明显,腰痛舒中、高剂量组在滑膜中的表达率较造模组下降,但在表浅层的表达率较造模组升高,TGF-β1表达在正常组有少量表达,造模后明显增加,腰痛舒高、中剂量组在滑膜中的表达减少。结论腰痛舒胶囊通过调节软骨bcl-2、p53以及软骨面、滑膜的PCNA和TGF-β1的表达,来延缓软骨细胞的凋亡,抑制滑膜的增殖。
Objective To observe the influence of Yaotongshu Capsules on the expressions of p53, bcl- 2, PCNA, TGF-β1 in the model of osteoarthritis (OA). Method To observe the expressions of p53, bcl-2, PCNA, TGF-β1 by the light microscope after immunohistochemical staining. Result There were no expressions of p53 and bcl-2 observed in the most samples of the normal group. After the model was established the sporadic expressions of p53 and bcl-2 were observed in cartilage transitional layer. After the administration of Yaotongshu Capsules the bcl-2 expression was increased gradually and p53 expression was decreased gradually. There was the sporadic PCNA expression observed in the normal group, and it was significant in synovial membrane in the model group. The PCNA expression in synovial membrane were decreased in groups with medium and high dose of Yaotongshu Capsules compared with that of the model group, but in surface layer it was increased. There was the sporadic TGF-β1 expression observed in the normal group, after the model establishment it was increased significantly and decreased in synovial membrane in the groups with high and medium dose of Yaotongshu Capsules. Conclusion Yaotongshu Capsules can delay the apoptosis of cartilage cells, inhibit the proliferation of synovial membrane, and promote cartilage plerosis through regulating the p53 and bcl-2 expressions of cartilage and the PCNA and TGF-β1 expressions of cartilage surface and synovial membrane.
