摘要
目的观察新一代血管紧张素转化酶抑制剂(ACEI)福辛普利(FOS)对体外培养的大鼠肾小球系膜细胞(GMC)产生的细胞外基质(ECM)成分层粘连蛋白(LN)的影响及FOS防治肾小球硬化的机制。方法按经典方法体外培养大鼠GMC,转种培养后分为对照组、脂多糖(LPS)组和LPS+FOS组。采用ELISA法测定GMC培养上清液LN含量,荧光半定量PCR方法检测LNmRNA的表达。结果LPS组GMC分泌LN蛋白及mRNA的表达明显高于对照组(P<0·05),LPS+FOS组GMC分泌LN蛋白及mRNA的表达较LPS组明显下降(P<0·05)。结论FOS从蛋白和mRNA两个水平对体外培养大鼠GMC产生的LN有明显的抑制作用,提示FOS抑制GMC产生ECM可能是延缓肾小球硬化的重要作用机制之一。
Objective To observe the effect of Fosinopril ( FOS), the new generation angiotensin - converting enzyme inhibitor ( ACEI), on expression of Laminin ( LN ), a factor of extracellular matrix ( ECM ), by rat glomerular mesangial cell (GMC) cultured in vitro ; to investigate the mechanism of FOS on prevention and treatment of glomerular sclerosis. Methods Rat GMCs were cultured in classic way. The cultured cell were divided into 3 groups ( 1 ) Control group, (2) LPS group, and ( 3 ) LPS + FOS group. LN concentration in GMC supernatant fluid was detected by ELISA; and LN mRNA expression was estimated by semiquantitative real - time RT PCR. Results LN secretion and mRNA expressions were obviously higher in LPS group than those in control group ( P 〈 0. 05 ) and decreased distinctively in LPS + FOS group compared with LPS group ( P 〈 0.05 ) . Conclusion FOS has obvious inhibited LN expression of rat GMC at both protein level and mRNA level, which suggest that the inhibition by FOS of components of glomerular extracellular matrix produced by rat glomerular mesangial cells is one of important mechanisms for prevention the development of glomeruloselerosis
