摘要
目的观察阿司匹林对大鼠脑缺血-再灌损伤24h和72h的脑保护作用。方法线栓法制作局部大脑缺血2h、再灌24h或72h模型,观察6m g.k-g 1及60m g.kg-1阿司匹林对脑损伤面积、水肿程度、死亡率、正常锥体神经元密度及脑组织病理形态的影响。结果C IR I 24h,脑损伤面积、水肿程度及死亡率均增加,正常锥体神经元密度下降,脑组织病理学异常改变。C IR I 72h时,与24h相比,脑损伤面积降低,水肿程度、死亡率、正常锥体神经元密度及脑组织形态学无明显改变。6m g.kg-1及60m g.kg-1阿司匹林明显缩小再灌注24h和72h引起脑损伤面积,降低脑水肿及死亡率,提高正常锥体神经元密度,改善脑细胞形态。结论阿司匹林对脑缺血-再灌损伤24h和72h均有明显脑保护作用。
OBJECTIVE This study, we design to investigate the protective effects of aspirin (ASA)on neurocytes after cerebral isehemia-reperfusion injury(CIRI) for 24 h or 72 h in rats. METHODS Right middle cerebral artery was occluded by inserting a thread through internal carotid artery for 2 h,and then reperfused 24 h or 72 h 6 and 60mg·kg^-1 doses of aspirin were ig administrated at reperfusion 0 h and 6 h,respectively. The brain injured area, cerebral edema, the mortality, the neuronal density and the brain morphology were estimated. RESULTS After CIRI 24h, the brain injured area, cerebral edema and the mortality were significently increased ,the neuronal density in injured regions was decreased. The brain morphology was abnormal. Compare with 24 h, the brain injured area was reduced after CIRI 72 h, and no significant change in cerebral edema, the mortality, the neuronal density in injured regions and the brain morphology was found after 72 h With using of 6 or 60mg·kg^-1doses of ASA, the brain injured area, and cerebral edema and the mortality were dramatically reduced, the neuronal density and the brain morphology were improved. CONCLUSION ASA has protective effect of neurocytes after CIRI either 24 h or 72 h.
