Expression and significance of Skp2,PTEN and p27 in human glioma

摘要 目的探讨skp2、PTEN和p27在胶质瘤中的表达及其意义,及其相互关系。方法用免疫组织化学方法检测33例胶质瘤和6例正常脑组织标本中Skp2、PTEN和p27蛋白的表达。结果Skp2在正常脑组织中表达不明显,在低级别胶质瘤中表达较少,在高级别胶质瘤中表达较多,两两比较差别有统计学意义(P<0.01)。PTEN和p27在正常脑组织中表达明显,在低级别胶质瘤中表达较少,在高级别胶质瘤中表达不明显,两两比较差异有统计学意义(P<0.01)。胶质瘤中skp2蛋白表达与p27蛋白表达呈负相关(r=-0.809,P<0.01),PTEN蛋白和p27蛋白的表达呈正相关(r=0.865,P<0.01)。Skp2蛋白和PTEN蛋白的表达呈负相关(r=-0.814,P<0.01)。结论在胶质瘤中PTEN的缺失突变导致Skp2在胶质瘤中表达升高,进一步促进p27的泛素化降解,使p27表达降低,失去对细胞周期的调控,从而促进胶质瘤的恶性进展和增殖。 Objective To investigate the significance and the expression of Skp2, PTEN and p27 in the human glioma, and their relationships. Methods The expression of Skp2, PTEN and p27 in 33 cases of human gliomas and 6 specimens of the normal brain tissue was detected by immunohistochemical technique. Results In normal brain tissue, groups Ⅰ,Ⅱand Ⅲ, Ⅳ, the positive expression level of Skp2 was 3.33 ± 1.21, 27.11 ± 9.29 and 59.53 ± 17.84 respectively, that of PTEN was 89.33 ± 6.89, 57.67± 14.47 and 8.13± 3.96 respectively, and that of p27 was 79.83± 10.11, 60.83± 14.04 and 13.07 ± 5.66 respectively. There were significant differences in the expression levels of Skp2, PTEN and p27 among the normal brain tissue, the gliomas of grades Ⅰ ,Ⅱ and the gliomas of grades Ⅲ, Ⅳ ( P〈0.01). Inverse correlation was found between protein of Skp2 and p27 in glioma, also between Skp2 and PTEN. There was a positive correlation between PTEN and p27. Conclusion The expression of Skp2 was increased due to the mutation of PTEN in gliomas. Therefore the degradation of p27 in the ubiquitinproteasome system was promoted. Skp2 may play an important role in the development of glioma by its promotive effect on decomposition of p27 and it may function as a critical component in the pathway of PTEN regulating p27 and cell proliferation in gliomas.
