钙通道抑制剂对人肝细胞钙池操纵钙通道电流的影响 被引量:2

Effects of 2-APB on store-operated Ca^(2+)channels currents in human hepatocytes

摘要 目的探讨钙池操纵钙通道(SOC)抑制剂(2-APB)对人肝细胞SOC电流(Isoc)的影响。方法应用全细胞膜片钳记录技术测定人肝细胞Isoc,并观察2-APB对该电流的影响。结果(1)培养的人肝细胞记录到Isoc,从其电流-电压(Ⅰ~Ⅴ)曲线可以看出该Isoc的反转电位为+3.0 mV左右。在钳制电位为-100 mV时,该Isoc为(-682.83±171.40)pA(n=8);(2)60、80μmol/L的2-APB分别使Isoc由给药前的(-682.83±171.40)pA下降至给药后的(-359.54±201.85)、(252.15±158.81)pA,给药前后差异有统计学意义(n=8,P<0.05或P<0.01);2-APB的抑制作用呈浓度依赖性增强,其EC_(50)为(63.36±9.57)μmol/L。结论2-APB对培养的人肝细胞Isoc具有显著的抑制作用。 Objective To study the effects of 2-APB on store operated calcium current (Isoc) in human hepatocytes. Methods Whole cell patch damp techniques were used to investigate the effects of 2-APB on Isoc in human hepatocytes . Results (1) 2-APB (20, 40, 60, 80 and 100) μmol/L produced a concentration-dependent decrease of Isoc with IC-60 value of (63.36 ± 9.57) μmol/L ( n = 8) ; (2) The peak value of Isoc in theⅠ-Ⅴ relationship was decreased by 2-APB 60 and 80 μmol/L from ( - 682.83 ± 171.40) before administration to ( - 359.54 ± 201.85) and (252.15± 158.81) ( n = 8, P〈0.05 or P〈0.01 vs control) respectively. But the reverse potential of Isoc occurred at voltage = 0 mV in all cells. Conclusion 2-APB has a protective effect on human hepatocytes, probably though inhibiting Isoc.
