摘要
目的探讨Notch-1和Notch-2在卵巢肿瘤的形成和转化中的表达特点及其潜在肿瘤生物学意义。方法2000-12-2003-06采用人类卵巢肿瘤组织芯片和免疫组织化学染色方法,在完全相同的实验条件下检测各类卵巢肿瘤Notch-1和Notch-2的表达和活化情况。结果Notch-1在50%(3/6)的浆液性囊腺瘤和33·3%(2/6)的粘液性囊腺瘤中有核易位现象;其中,16·7%(1/6)和33·3%(2/6)属于明显易位(++/+++)。在交界性肿瘤中的核易位无论是频率(20%;1/5)还是易位的阳性程度均明显减低,而这种趋势在各类卵巢恶性肿瘤中亦可见到。Mann-WhitnoyU检验显示:Notch-1的核易位在良性和恶性肿瘤之间差异有统计学意义(P<0·05)。Notch-2普遍存在于各类卵巢上皮肿瘤中;其表达水平和核易位情况与肿瘤的良恶性之间无关。结论Notch-1的表达异常和核易位出现在卵巢上皮的恶性转化的过程,它可能是卵巢肿瘤恶变风险和不良预后的生物学标志物。
Objective To study the expression and nuclear translocation of Notch - 1 and Notch - 2 in different ovarian tumors and to analyze its clinical biological significance in tumor transformation and progression. Methods The ovarian tumor array - based immunocytochemical staining was performed to check the expression patterns and intracellular distribution of Notch - 1 and - 2 in ovarian tumors under the same experimental condition. Results The rates of Notch - 1 nuclear translocation were 50% (3/6)and 33, 3% (2/6)in serous and mucinous cystadenoma respectively, among which 16.7% (1/6) and 33.3% (2/6) showed more distinctively ( + +/+ + + ). In contrast, the borderline and malignant tumors exhibited downregulated Notch - 1 expression as well as reduced and even diminished nulear translocation (P 〈 0.05 ). Notch -2 expression and nuclear translocation could be observed either in benign tumors or in borderline and malignant ones ( P 〉 0.05 ). Conclusion The aberrant Notch - 1 but not Notch - 2 expression occurs in the transformation phase of ovarian tumors, indicating its value in the evaluation of transformation risk and poor prognosis of ovarian tumors.
