摘要
[目的]研究Flt-3配体(FL)及血小板生成素(TPO)在无血清培养体系中对人脐带血CD 34+细胞体外增殖的影响.[方法]采用无血清液体培养法,分别用FL,TPO,FL加TPO作用于体外短期培养脐带血CD 34+细胞,利用其计数及集落分析方法评价增殖效果.[结果]分别用FL,TPO,FL加TPO刺激增殖后,人脐带血细胞总数分别增加2.50,5.00,10.10倍,FL加TPO组细胞总数明显高于FL,TPO组;CD 34+细胞分别增殖0.90,1.26,1.61倍;集落总数分别增加2.07,2.27,7.37倍,造血祖细胞CFU-GM产率分别提高3.53,2.40,9.95倍,FL加TPO组产率高于其他组,BFU-E产率分别提高1.45,2.25.6.90倍.[结论]在短期无血清液体培养体系中,FL,TPO对人脐带血CD 34+细胞均具有增殖作用,且两者具有协同作用.
OBJECTIVE To investigate the expansion effects of Flt-3 ligand (FL) and thrombopoietin (TPO) on human umbilical cord blood CD 34 + cells. METHODS CD 34 + cells from cord blood were cultured with FL and TPO alone or combination with the two cytokines in serum-free liquid culture system. The expansion effects were evaluated with cell count, CD 34 + cells and colony assay. RESULTS The number of nucleated cells increased 2.50 , 5.00 and 10.10 fold in FL, TPO and FL + TPO group respectively, the cell counts of the combination group were higher than control group significantly. Moreover, the number of nucleated cell in FL+ TPO group was significantly higher than that of cells in FL or TPO alone. After expansion, the CD 34 + cells increased 0.90,1.26 and 1.61 fold in FL. Clony forming cells (CFC) which remarkable enhanced in all three groups after culture, were increased 2.07, 2.27 and 7.37 fold. The number of CFU-GM in FL, TPO and FL+ TPO group was significantly higher, increased 3.53, 2.40 and 9.95 fold after culture. The yield of BFU-E was increased 1.45 fold and 2.25 fold in FL and TPO group, which enhanced 6.90 fold in FL + TPO group. CONCLUSION Both FL and TPO can expand CD 34 + cells from cord bloodfairly in short term serum-free liquid culture, the comnation with FL and TPO showed remarkable synergistic effect.
