摘要
以鲁北冬枣一年生枣头刚萌发的顶芽、嫩枝枣头先端和一年生枣头上未萌发的侧芽及带芽茎段为试材制备外植体,进行组培快繁试验。结果表明,以嫩枝枣头先端做培养材料较适宜,成活率可达22.2%,萌发生长较快,且操作容易。最佳分化增殖培养基为MS+6-BA2.0mg/L+KT0.5mg/L+2,4-D0.2mg/L;最佳生根培养基为1/2MS+IAA0.5mg/L+IBA1.0mg/L。移栽时注意遮荫和控制温度,成活率达90%。
Tissue culture of winter jujube ＇ Lubeidongzao＇ （ native to the north of Shandong Province） with different tissue parts as explants manifested that the shoot tip got the highest survival rate（22. 2% ） ,it germinated and grew more quickly,and was easy to be operated. The optimum differentiation and multiplication medium was MS ＋6- BA 2. 0rag/L ＋KT 0. 5mg/L ＋ 2,4 - D 0. 2mg/L;the optimum rooting medium was 1/2MS ＋ LAA 0. 5mg/L ＋ IBA 1.0mg/L. The transplanting survival percentage could be obtained by 90% under the shading and suitable temperature conditions.
