胎儿骨髓间充质干细胞中SSEA-4阳性表达细胞的分离和检测 被引量:5

摘要 为了验证成体干细胞中全能性干细胞存在的可能性,以SSEA-4抗原为筛选标志,利用免疫磁珠筛选技术从胎儿来源的骨髓间充质干细胞(fetal mesenchymal stem cells,fMSCs)中筛选并培养得到SSEA-4表达阳性的fMSC-SSEA-4细胞,从形态学、生长曲线、表面标志及细胞周期、核型分析等方面检测其生物学性状,并通过Oct-4及SSEA-4抗体的表达及致瘤性等方面分析其全能性状.同时观察了fMSC-SSEA-4细胞的诱导分化能力.结果发现,fMSC-SSEA-4细胞的形态学表现、细胞周期和生长曲线检测与fMSCs无明显差异;fMSC-SSEA-4细胞Oct-4和SSEA-4抗体表达阳性,并且表现为正常的二倍体核型,无明显的致瘤性状.在不同的诱导条件下,fMSC-SSEA-4细胞可向成骨细胞、脂肪细胞和神经元样细胞分化.实验结果提示,fMSCs中存在有全能干细胞性状的一群细胞,从而为进一步深入研究和更好地应用fMSCs提供了理论依据.
