摘要
目的:探索神经干细胞与生物活性材料壳聚糖或明胶体外共培养构建的工程化组织的最佳体内移植时间。方法:从胎龄14~16 d的Wistar大鼠大脑分离培养神经干细胞,以105 cells/ml密度种植于壳聚糖、明胶膜表面,每天观察神经干细胞的分化情况。结果:培养获得的神经干细胞具有自我更新和多向分化潜能,接种于壳聚糖和明胶膜后24 h均贴附牢固,48 h后均有细胞分化。结论:神经干细胞与壳聚糖或明胶体外共培养构建的工程化组织的最佳移植时间为神经干细胞接种后24~48 h之间。
Objective: To explore the optimal implantation time for engineering tissue based on neural stem cells (NSCs) and biomaterials chitosan or gelatin film in vitro. Methods: NSCs were dissociated and cultured from the brain of 14-16 days old embryonic Wistar rats. NSCs were seeded on the surface of Chitosan and gelatin film respectively at a density of 105cells/ml. The attachment and differentiation of NSCs were observed daily. Results: Cultured NSCs had the capacity of self-renewing and multi-potential differentiation. NSCs attached firmly to the surface of both chitosan and gelatin films 24 hours after their seeding and differentiated cells appeared 48 hours later. Condusion: The optimal implantation time for NSCs based engineering tissue ranges from 24 hours to 48 hours after NSCs were seeded.
