摘要
目的探讨人类重组拉-巨噬细胞集落刺激因子(rhGM—CSF)对照射狗脾脏及淋巴结修复作用的影响.方法雄性Beagle狗14只.均双例髂骨屏蔽,其余部位用60Coγ射线照射.除对照组外.治疗组股静脉注射给药.连续20d,低剂量组为10μg·kg-1·d-1.高剂量组20μg·kg-1·d-1.30d时处死动物.用HE和AgNOR染色观察脾脏和淋巴结结构的变化,并对脾小体和淋巴滤泡面密度及数密度进行定量分析.结果低剂量组淋巴结和脾脏组织结构中等程度恢复;淋巴细胞增殖不十分明显.而高剂量组的牌及淋巴结基本恢复正常,淋巴细胞增殖明显高于对照组.结论rhGM—CSF可促进照射后构狗脾脏及淋巴结形态与功能的恢复,其恢复程度与该因子的剂量有关.
Objective TO explore the effects of rhGM-CSF on the reperation of spleen and lymph node in irradiated dog.Methods 14 male dogs were shielded on iliums, the other positions exception the ilium were irradiated by As ray. rhOM-CSF was injected per vein till 20 days in low dosage group μg·kg-1.d-1 and high dosage group ug·kg-1.d-1.At the 30th day, the spleen and lymph node were made into slides (H-E and AgNoR staining), the changes of structure of spleen and lymph modes were observed under the microscope. Area density and number density of splenic corpuscle and lymph follice were analysed in quantitative method.Results The structure of spleen and lymph node in low dosage rhGM-CSF group recovered moderately, lymphocyte proliferation was not very apparent, however in high dosage group, the structure have basically recovered to normal level,the proliferation rate of lymphocyte was apparently higher than that of control group (P<0.01).Conclusion It is suggested that rhGM-CSF could promote the recovery of morphosis and function of irradiated mmune organs and the recovery degrees are related to the dosage of rhGM-CSF.
