Selection and Sequence Analyses of High Affinity Binding Peptides for Ni^(2+)

摘要 利用噬菌体随机十二肽库对金属N i2+进行了结合肽筛选,经5轮生物淘洗、噬菌体扩增和DNA测序,获得8条多肽序列,分析发现其中富含组氨酸(4~6个/肽).结果表明,组氨酸的存在对蛋白质和金属N i2+的结合起着关键作用. Phage random 12-peptide library was used for selecting peptide binding for Ni^2+ . After five rounds of biopanning, phage amplification and DNA sequencing, a group of 12 - peptides were obtained from the library. The sequences of these DNA coding peptides were sequenced and identified, containing four to six histidine residues. The results showed histidine residues were very important in the binding peptides for Ni^2+. Fig 1, Tab 1, Ref 11
