异搏定对肾癌细胞多重耐药性的逆转作用 被引量:2

Overcoming the Multidrug Resistance in Renal Cell Carcinoma of RCC-925 Cell Line in Vitro by Means of Verapamil

摘要 通过肾癌细胞的体外实验,应用MTT方法,观察了化疗致敏剂搏定对细胞毒药物长春新碱和阿霉素抗RCC-925肾癌细胞效应的体外增效作用.结果发现:RCC-925肾癌细胞系对长春新碱高度耐药,对阿霉素低度耐药,半数致死浓度分别为14726.4μg/ml和1.559μg/ml。异搏定1.25μg/ml即可显著提高长春新碱和阿霉索的细胞毒性,增效幅度随异搏定浓度的升高而增强,增效倍数对长春新碱为1.3×104~1.5×105倍,对阿霉素为3.5~25.1倍。提示异搏定对RCC-925肾癌细胞的多重耐药性具有显著的逆转作用. Using MTT method, the effects of verapamil on the enhancement of cytotoxicity of Vincristine(VCR)and Adriamycin (ADM) against renal cell carcinoma cell line RCC-925 in vitro were investigated. RCC-925cell line showed high resistance to VCR and moderate resistance to ADM. The LC50. was 14726. 4μg/ml for VCRand 1. 559μg/ml for ADM,suggesting that RCC-925 cell line have the natural multidrug resistance. Verapamil atthe concertration above 1. 25μg/ml could significantly enhance the cytotoxicity of VCR and ADM,the extent of enhancement was correspondingly increased as the dose was increased and the multiplication was from1. 3 × 104 to 1. 5 × 105 in VCR and 3. 7 to 25. 1 in ADM. The results suggested that verapamil dramatically reversemultidrug resistance in RCC-925 cells.
