摘要
目的探讨在糖耐量受损(impairedglucosetolerance,IGT)期大鼠血清脂联素(adiponectin,APN)与瘦素(leptin)的变化及其与冠状动脉病变的关系。方法Wistar大鼠80只,随机分为对照组和实验组,实验组以高脂膳食饲养,诱导IGT模型,检测血清APN、leptin、FINS等指标,并在光镜下观察冠状动脉的病理变化,测定病理切片上冠状动脉内中膜的厚度(IMT)。结果IGT组大鼠血清APN在第8周时较正常对照组下降(P<0.05);leptin第8周时较正常对照组显著上升,差异有统计学意义(P<0.05);左冠状动脉内中膜厚度在12、20周均值分别达(16.5±1.8)μm、(26.7±1.7)μm,较正常对照组厚度明显增加(0.01<P<0.05),平滑肌排列紊乱并向内膜下迁移。结论血清脂联素下降、leptin升高与IGT期冠状动脉病变发生显著相关,提示血清低ANP、高leptin参与了心血管病的发生与发展,其中ANP发挥保护性作用,而leptin发挥损伤性作用。ANP水平下降,leptin升高可能成为预测糖尿病前期冠状动脉病变的一个重要指标。
Objective To explore the role of adiponectin and leptin in the development of coronary artery lesion in IGT rats.Methods Wistar rats were randomly divided into normal control group and enriching-fat-diet feeding control group.At 8,12 and 20 weeks,the early changes of the coronary artery were examined by light microscope.Computerized image processing was employed to measure the coronary artery intimal medial thickness (IMT).Plasma adipenectin and leptin were measured by ELISA.Result The early stages of coronary lesion occurred in IGT 12 weeks.The decrease of plasma adiponectin were observed at 8 weeks.The coronary artery intima-media thickness was correlated positively with the adipenectin.Conclusion The decrease of adipenectin occurred before visible and measurable coronary artery pathological changes.Adipenectin may play an important role in the progression of IGT coronary artery lesion.It can be used as an early figure to predict IGT coronary artery.
