摘要
以重组TgoDNA聚合酶为研究对象,检测了其最适扩增条件。结果表明:该重组酶的最适pH为8.75,最适Mg2+浓度为2.0 mmol/L,最适延伸温度为71.9℃。利用该重组酶成功扩增了质粒pUC19、野大麦Na+/H+逆向运转蛋白基因、拟南芥Timeless基因、小鼠Cx37基因和人CYT2C9基因。
The optimum amplification conditions of the recombined Tgo DNA polymerase were examined in this study. The results revealed that the optimum PH of the enzyme was 8.75, the optimum concentration of Mg^2+ was 2.0 mmol/L, and the optimum elongating temperature was 71.9 ℃. Plasmid pUC19, Na^ +/H^+ reversed transporting protein gene of wild barley, Arabidopsis Timeless gene, mouse Cx37 gene and human CYT2C9 gene were amplified successfully using this recombined enzyme.
