摘要
目的:探讨共刺激分子B71和B72在大鼠肝脏热缺血伤再灌注损伤模型中的表达情况及其免疫学意义。方法:将雄性Wistar大鼠随机分为3组:A组(缺血30min再灌注24h)、B组(缺血60min再灌注24h)、C组(假手术组)。模型制备参考Ohmorid的方法。分别取模型之肝左叶,采用实时反转录聚合酶链反应(PTPCR)检测B71和B72在3组肝左叶组织中mRNA表达情况。结果:B71和B72mRNA在C组以极低水平表达,而在A组、B组表达却明显增多(P<0.01),且B组高于A组(P<0.05)。结论:热缺血再灌注之肝脏通过上调B71和B72的表达增加了肝脏的免疫原性。
Objective:To Investigate the expression of costlmulatory molecule B7-1 and B7-2 by warm ischemia/reperfusion(1/R)injury In rat liver and the meening of ImmunolosY. Methods : Male Wistar rats were randomly dlvicted Into 3 groups:A(ischemia 30 mins, referfusion 24h), B(ischemia 60 mins,referfusion 24h),C(sham operation group). A model of hepatic warm I/R in rats was establlshed according to the method of Ohmorid. The expression of B7-1 and B7-2 mRNA was analyzed by real-time reverse transcription polyrnerase chain reaction(RT-POR), Results :Oompared with the sham operation group,the expression of B7-1 and B7-2 mRNA increased sisnificently In the rats unclersoing I/R procedure(P〈0.01).Conclusion:Warm ischemia/reperfusion injury may increase the Immunosenicity of the liver by upresulating the costimulatory molecule BT-1 ancl B7-2.
