摘要
对新生仔兔胰腺干细胞的分离、培养方法进行了研究,并检测了其横向诱导分化为心肌、成骨、神经细胞的潜能。结果表明,分离到的新生仔兔胰腺干细胞为上皮样细胞,在传代过程中该细胞的生长特性未发生改变,并始终表达导管源胰腺干细胞的标志CK-19,PDX-1;新生仔兔胰腺干细胞经诱导可分化为心肌、神经和成骨细胞,经免疫组化染色鉴定显示,心肌样细胞表达-αactin、神经样细胞表达NF-200而不表达GFAP、成骨样细胞茜素红染色呈阳性。证明该细胞具有多潜能性,是一种亚全能干细胞。
This research investigated the isolation and culture of neonatal rabbit pancreatic stem cells, examined the capacity of differentiation into myocardial-like cells ,neurons osteoblasts. The results demonstrated that the cell isolated was epithelium cell ,whose growth characteristic did not change in the process of subculture,and expressed the markers of ductal pancreatic stem cell of CK-19 and PDX-1. This cell can be induced to differentiate into myocardia, neurons,osteoblasts, It's positive for α actin and NF-200 but negative for GFAP by the method of immunohistochemical staining and Alizarin Red staining. The result proves that this cell is a type of subtotipotent stem cell resided in pancreas.
