A method of using plasmid PUC18 directly for expression cDNA gene in E. coli was reported. The human protein disulfide isomerase cDNA (PDI cDNA)was inserted into the LacZ sequence of plasmid pUC18 to construct the PDI expression vector and expressed in E. coli as a soluble protein. The PDI expression level was about 24% of total cell protein, and the PDI isomerase activity of the bacteria lysis supernant was 12 U/L.
