摘要
目的:观察电针治疗对大鼠脊髓损伤后神经细胞再生修复过程中Fas和半胱氨酸天冬氨酸蛋白酶3表达的影响,并与甲基强的松龙进行阳性对照。方法:实验于2003-04/2004-06在佳木斯医学院中心实验室进行。①将健康成年Wistar大鼠64只单纯随机分为模型组、电针组、甲基强的松龙组及假手术组4组(n=16),除假手术组外,其他3组大鼠采用改良的Allen’s捶击法(50g·cm)制成大鼠T10脊髓损伤模型,假手术组仅切除椎板,不损伤脊髓。②电针组在损伤后即刻取督脉上的大椎和命门两(进行治疗,电针频率2Hz,疏密波形,强度2.5mA,以针刺处肌肉轻微抖动为宜,持续30min。甲基强的松龙组在损伤后即刻尾静脉内注射30mg/kg甲基强的松龙。模型组和假手术组不做治疗。③各组分别于术后6,24h各处死8只。应用免疫组织化学方法及图像定量分析法观察各组脊髓损伤早期Fas和半胱氨酸天冬氨酸蛋白酶3表达。结果:经补充后61只大鼠进入结果分析。①脊髓损伤后6h:电针及甲基强的松龙组Fas及半胱氨酸天冬氨酸蛋白酶3阳性细胞数少于假手术组(P<0.05),灰度值高于假手术和模型组相(P<0.05,0.01)。②脊髓损伤后24h:电针及甲基强的松龙组Fas及半胱氨酸天冬氨酸蛋白酶3阳性细胞数少于假手术组和模型组(P<0.05),灰度值高于假手术和模型组(P<0.05,0.01);但电针与甲基强的松龙组组间差异不显著(P>0.05)。结论:电针可使Fas及半胱氨酸天冬氨酸蛋白酶3的表达下调,从而抑制细胞凋亡,保护神经细胞,对损伤脊髓的修复有积极的治疗作用,其效果与甲基强的松龙相似。
AIM: To observe the expressions of Fas and Capase-3 during the regeneration and repairing process of nervous cells in rats after acute spinal cord injury (SCI) treated with electro-acupuncture (EA), and conduct positive comparison with methylprednisolone (MP).
METHODS: The experiment was done in the central laboratory of Jiamusi Medical College from April 2003 to June 2004.①Sixty-four healthy adult Wistar rats were simply divided into model group, EA group, MP group and sham operation group with 16 rats for each. Models of T10 SCI were induced by modified Allen's method (50 g·cm) in rats except the sham operation group, in which the rats were only given vertebral lamina resection without injuring spinal cord.②EA group: Immediately after SCI, Tachui (DU 14) and Miagmen (DU 4) were pricked at sparse-dense waveform of 2.5 mA intensity and 2 Hz frequency for 30 minutes. It would be suitable if the muscles of EA treatment shivered gently. MP group: 30 mg/kg MP was injected intravenously into rat tails instantly after SCI. There was no treatment in model group and sham operation group.③Eight rats were executed respectively at postoperative 6 hours and 24 hours. The expressions of Fas and Caspase-3 in early SCI were observed by immunohistochemical analysis and image quantitative analysis.
RESULTS: A total of 61 rats were involved in the result analysis after supplement. ①Six hours after the operation, the expressions of Fas and Caspase-3 decreased in EA group and MP group compared with sham operation group (P 〈 0.05), and gray value was higher than that in sham operation group and model group (P 〈 0.05, 0.01). ②Twenty-four hours after the operation, EA group and MP group demonstrated lower expressions of Fas and Caspase-3 than sham operation group and model group (P 〈 0.05), whereas the gray value was higher than that of sham operation group and model group (P 〈 0.05, 0.01); But there were insignificant differences between EA group and MP group �
