MR影像学评价VEGF对兔局灶性脑缺血再灌注损伤的有效治疗时间窗 被引量:4

Study on Therapeutic Window of Opportunity for VEGF Following Focal Cerebral Ischemia Reperfusion Injury in Rabbits by MR Image Technology

摘要 目的:探讨MR影像学评价血管内皮细胞生长因子对家兔局灶性脑缺血再灌注损伤的有效治疗时间窗。材料和方法:采用兔大脑中动脉阻断局灶性脑缺血再灌注模型,分别在缺血2h再灌注损伤后0h、1h、3h、6h和12h应用微量进样器将VEGF立体定向导入梗死灶周,于再灌注72h,应用MR影像学、TTC染色和流式细胞术评价梗死体积、灶周缺血半暗带凋亡率、ADC值比率(ADCR),评价神经功能缺损。结果:缺血再灌注0h和1h灶周给予VEGF,MRI测得的梗死体积分别为(367.0±15.7)mm3和(393.0±19.6)mm3与对照组(468.6±29.7)mm3相比差异有显著性(P均<0.01);3h、6h和12h组梗死体积为(437.0±24.6)mm3、(444.0±29.5)mm3、(449.0±30.5)mm3与对照组相比差异没有显著性(P>0.05)。TTC染色测得的梗死体积与MRI上相一致。同时1h内用药神经功能缺损评分明显减少,凋亡率明显下降,再灌注3h后给药,则无明显作用。结论:VEGF对兔局灶性脑缺血再灌注损伤的有效时间窗在再灌注损伤1h内,MR影像学检查可作为定量评价基因疗效的可靠指标。 Purpose: To study the therapeutic window of oppourtunity for VEGF following focal cerebral ischemia reperfusion injury in rabbits and the effectiveness was evaluated by advanced MR imaging technology. Materials and Methods : Focal cerebral ischemia 2h / reperfusion 72h model in rabbits was induced by transient occlusion of middle cerebral artery(MCA) for 2h and reperfusion for 72h. The VEGF was ad- ministered at Oh, lh, 3h, 6h and 12h after reperfusion respectively. We measured infarct volumes, neural cell apoptosis and ADCR in the perifocal regions of cerebral ischemic and neurological deficit score 72h after reperfusion.Results : When administered VEGF at Oh and lh after reperfusion, Infarct vol- umes, neurological deficit score and the percentage of apoptotic cell were significantly lower than that in control group (P 〈 0.01) . There was no difference when administered VEGF at 3h after reperfusion. Conclusion : The therapeutic window of opportunity for VEGF following focal cerebral ischemia/reper- fusion injury in rabbit is not more than lh after reperfusion. MRI could be quatitatively evaluated the effect gene therapy in vivo.
