摘要 目的:建立高效液相色谱法-蒸发光散射检测器测定脂肪乳注射液中磷脂酰胆碱(PC)和磷脂酰乙醇胺(PE)的含量。方法:采用 Kromasil 100-5SIL 硅胶色谱柱(250mm×4.6mm,5μm);以正己烷-异丙醇(3:4)为流动相 A,正己烷-异丙醇-水(3:4:0.75)为流动相 B;流速为1.5mL·min^(-1);蒸发器温度100℃;雾化气:氮气,喷气流速为1.7L·min^(-1)。结果:PC的线性范围为0.08~0.56mg·mL^(-1),r=0.9982(n=7);PE 的线性范围为0.04~0.16mg·mL^(-1),r=0.9978(n=7),PC 和PE 的加样回收率分别为98.8%,99.2%,99.0%和98.6%,98.9%,98.2%;RSD 分别为0.1%,0.1%,0.2%和0.1%,0.2%,0.1%。结论:本法操作简便快速,重复性好,灵敏度高,准确,专属性高。 Objective: To develop an HPLC - ELSD method for determination of phosphatidyl choline ( PC ) and phosphatidyl ethanolamine (PE) content in the medium and long chain fat emulsion injections. Method:The method adopted Kromasil 100 -5SIL column(250 mm ×4. 6 mm,5 μm) with n-hexane-/so-propanol(3: 4) as mobile phase A, n-hexane -/so-propanol - water( 3: 4: 0. 75 ) as mobile phase B, 1.5 mL ·min^-1 as flow rate, 100 ℃ as inlet heater, 1.7 L ·min^-1 as N2 flow rate. Result: The linear range for the content of PC was 0.08 - 0. 56 mg·mL^-1 ,r =0. 9982(n =7) and for PE was 0. 04 -0. 16 mg ·mL^-1 ,r =0. 9978(n =7). The recoveries of PC and PE were 98.8% ,99.2% ,99. 0% and 98.6% ,98.9% ,98.2% ,respectively. The RSDs were 0. 1% , 0. 1% ,0. 2% and 0. 1% ,0. 2% ,0. 1%, respectively. Conclusion: The method is simple, rapid, reproducible, good sensitive, accurate and is suitable for the determination of PC and PE.
