全自动多通道毛细管区带电泳法在血清蛋白分析中的临床应用 被引量:17

Automated capillary zone electrophoresis for analysis of human serum protein in clinical application

摘要 目的 用毛细管区带电泳(CE)技术进行血清蛋白检测结果与琼脂糖凝胶电泳(AGE)技术的检测结果进行比较分析以探讨CE的临床应用.方法 使用全自动毛细管电泳仪(法国Sebia公司的4.51版本Capillarys(R)电泳仪)及相应试剂,在7 kV电压、35.5℃、pH 10的缓冲液条件下,在15.5 cm×25.0 μm硅毛细管内进行血清蛋白电泳,用200 nm波长检测各种蛋白质的百分比浓度.结果 与AGE的结果比较相关性,进行CE的精密度、线性检测及干扰实验.CE和AGE比较,白蛋白、α1-球蛋白、α2-球蛋白、β-球蛋白及γ-球蛋白的相关系数分别为0.929、0.924、0.841、0.789和0.926,P<0.001相关性好,无统计学意义.蛋白质含量3个水平不同标本的批内、批间结果,白蛋白、α1-球蛋白、α2-球蛋白、β1-球蛋白、β2-球蛋白和γ-球蛋白的变异系数(CV)值均小于10.08%精密度好.血清总蛋白浓度在71.10~11.16 g/L范围内各种蛋白线性关系良好,白蛋白最低检出量为6.87 g/L,γ-球蛋白为1.78 g/L.血清总胆红素含量≤144.7 mmol/L、甘油三酯含量≤8.18 mmol/L时对CE结果无干扰,纤维蛋白原和血红蛋白对CE结果有干扰.结论 CE技术是一种灵敏度高、精确度好的血清蛋白检测技术,随着设备自动化的实现,分析操作将更简便、快速,适于临床化学实验室使用. Objective The automated capillary zone electrophoresis (CE) is used for human serum protein analysis and compared with the results of agarose gel eleetrophoresis (AGE) for human serum protein analysis. Discuss its' practical application in clinical. Methods With the Capillarys β1 -β2 + reagent set, proteins in serum were separated at 7 kV for 4 min in 15.5 cm × 25. 0 μm fused-silica capillaries (n = 8) at 35. 5℃ in a pH 10 buffer with online detection 200 nm. Results Capillary zone eleetrophoresis compared with agarose gel eleetrophoresis ( AGE; Hydrasys -Hyry ; Hydragel protein ( e ) 15/30/54 reagent set;Sebia) in precision, relatives, linearity and potential interferences were analyzed. A samples without Para-protein ( n = 165 ) were performed relation test. The coefficient of relation between CE and AGE is 0. 929 for albumin, 0. 924 for for α1-globulin, 0. 841 for α2-globulin, 0. 789 for β-globulin, 0. 926 for γ- globulin. There was no significant difference (P 〈0. 001 ). The three different level of protein concentration which were selected perform within-run or between-run for 24 times, coefficient variation (CV) were ≤ 3.51% for albumin, ≤10. 08 for α1-globulin, ≤6. 48% for α2-globulin, ≤5.6% for β1-globulin, ≤9. 56 for β2-globulin and 〈 4.07% for γ-globulin. Within the concentration of the total protein is 71.10-11.16 g/ L, the liner of all kind of bands is better. The lowest examination ratio of albumin and γ-globulin separately is 6. 87 g/L and 1.78 g/L. There is no affect when the concentration of total bilirubin in serum under 144. 7 mmol/L and concentration of TG under 8. 18 mmol/L. There is effect when concentration of hemoglobin is higher than 0. 75 g/L. The fibrinogen affected the results of multi-capillary zone eleetrophoresis. Conclusion The automated multi-capillary zone eleetrophoresis (CE) is a reliable,sensitive and faster method for serum protein analysis. It is easy to operate with combining advantages of full a
