银杏叶总黄酮对糖尿病大鼠视网膜细胞凋亡相关基因表达的影响 被引量:10

The influence of total Ginkgo flavonoids on the expression of apoptosis-related genes in retinas of diabetic rats

摘要 目的探讨银杏叶总黄酮(FG)对视网膜细胞凋亡相关基因表达的影响。方法48只SD大鼠随机分为正常对照(NC)、糖尿病对照(DC)、及不同剂量FG(L-FG1、L-FG2、M-FG、H-FG)治疗组,腹腔注射链脲佐菌素(STZ)诱发大鼠糖尿病。第5周起,各治疗组分别给予FG20、50、100、200mg/(kg·d)灌胃,共8周。采用免疫组化法检测各组大鼠视网膜细胞中Bax和Bcl-2基因表达强度,利用生物图像分析软件进行分析。结果Bax和Bcl-2在DC组视网膜血管和神经细胞较NC组免疫强度增强;在各治疗组的强度较DC组减弱,其中M-FG、H-FG组与DC组之间差异有显著性;各治疗组与NC组比较,其表达仍有增强,L-FG1、L-FG2组与NC组之间差异有显著性。结论FG可以影响视网膜细胞凋亡相关基因Bax和Bcl-2的表达,并可能由此影响糖尿病大鼠视网膜细胞的凋亡。 Objective To study the expression of the apoptosis-related genes ( Bax, Bcl-2) in retinas of diabetic rats before and after treatment with total Ginkgo flavonoids( FG), and evaluate the functions of FG on the expression. Methods Forty-eight SD rats were randomly divided into six groups of normal control ( NC), diabetic control ( DC), FG treatment( L-FG1, L-FG2, M-FG, H-FG) group. Diabetic models were induced by streptozotocin(STZ). After giving FG at 20,50,100,200 mg/( kg · d) to the treatment groups for 8 weeks, immunohistochemical (ABC) method was applied to monitor the expression, and then the results were analyzed using organism picture analysis system. Results The positive reaction intensity of both Bax and Bcl-2 was stronger in DC, L-FG1, L-FG2 than that in NC, while weaker in M-FG, H-FG than in DC. Conclusion FG can affect the expression of Bax and Bcl-2 in retinas, which may be related to apoptosis of retinal cell in diabetic rats.
