摘要
分别以番茄东农704、东农708、东农709三品种的子叶、茎段及菊花30、33、34的愈伤组织为材料,用不同浓度的潮霉素进行敏感性试验.结果表明,随着潮霉素浓度的升高,番茄子叶、茎段的出愈率下降,菊花愈伤组织的褐化率增加.在以潮霉素作为抗性选择标记时,选择压力番茄以15mg/L为宜,菊花3品种潮霉素的选择压力以15-20mg/L为好.
The sensibility to different concentration of Hygromycin was researched by taking cotyledons and petiol segments of tomato Dongnong 704, 708, 709 and callus of chrysanthemum 30, 33, 34 as material respectively. The results were as follows, frequency of callus formation were decreased in tomato Cotyledons and Petiol segments, and frequency of browning were increased in callus of chrysanthemum with the increasing concentration of Hygromycin. The ideal selecting pressure of Hygromycin is 15mg/L in tomato, and 15-20mg/L in chrysanthemum when Hygromycin is used as selection marker.
