摘要 目的:探讨自体外周血干细胞动员时,恶性血液病患者的骨髓及外周血CD34+和T细胞亚群的动态变化特点。方法:16例拟行自体外周血干细胞移植患者,在应用化疗+G-CSF方法动员外周血干细胞期间,以流式细胞仪测定骨髓及外周血CD34+和T细胞亚群动态变化情况。结果:化疗后应用G-CSF 48 h后,骨髓中CD34+细胞开始明显增加(P<0.01),在应用G-CSF 72 h后,外周血中CD34+细胞开始明显增加(P<0.01),96 h后,两者达峰值,较未应用G-CSF时差异显著(P<0.01)。T淋巴细胞亚群比例处于倒置状态且变化不明显(P>0.05)。结论:恶性血液病患者在应用化疗+G-CSF方法动员外周血干细胞时,其骨髓及外周血CD34+细胞逐渐增加,于96 h后达高峰,T细胞亚群变化不明显。 Objective:To study the dynamic changes of CD34^+ cells and T lymphocyte subsets in bone marrow or peripheral blood during peripheral blood stem cell mobilization by chemotherapy and G - CSF injection. Methods: Flow cytometry was used to detect CD34^+cells and T lymphocyte subsets in 16 patients treated with chemotherapy and injected with G - CSF for peripheral blood stem cells mobilization. Results: CD34^+cells in the bone marrow and peripheral blood increased obviously after injecting G - CSF for 48 h and 72 h respectively. The highest percentages were achieved 96 h after this procedure in both of them. There were obvious difference between before and after G - CSF injection ( P 〈 0.01 ). CD4/CD8 ratio was inverted in both bone marrow cells and peripheral blood cells, but there were no obvious changes(P 〉 0.05 ). Conclusion: CD34^+ cells in bone marrow or peripheral blood increase gradually after chemotherapy and G - CSF injection, and can reach the highest level 96 h after this procedure. T lymphocyte subsets keep stationary.
