摘要
目的探讨生存素蛋白在贲门癌及癌前病变组织中的表达及其与细胞凋亡的关系。方法应用免疫组化过氧化物酶标记的链霉卵白素(SP)法检测贲门炎(20例)、贲门不典型增生(Ⅰ级20例、Ⅱ级16例、Ⅲ级6例)和贲门癌(67例)组织中生存素蛋白的表达;应用DNA聚合酶和末端脱氧核糖核酸转移酶标记DNA链断端的核酶(TUNEL)法检测以上贲门组织中的凋亡细胞,计算凋亡指数(AI)。结果①从贲门炎到不典型增生(Ⅰ、Ⅱ、Ⅲ级)到贲门癌,随着病变的加重生存素蛋白表达率呈逐渐增高趋势,分别为0.0%、30.0%、25.0%、83.3%和61.2%;凋亡指数则呈逐渐下降趋势,分别为(15.15±6.97)%、(14.84±6.66)%、(14.33±5.34)%、(9.31±3.96)%和(6.47±2.82)%。②贲门癌组与Ⅲ级不典型增生组相比生存素蛋白表达差异无统计学意义(P>0.05),与其余各组比较表达差异有统计学意义(P<0.01)。贲门炎组、贲门Ⅰ、Ⅱ级不典型增生组间生存素蛋白表达差异无统计学意义(P>0.05)。③生存素蛋白表达与凋亡指数呈负相关(rs=-0.912,P<0.05)。结论生存素蛋白表达上调发生在贲门癌变过程的早期阶段,并通过抑制细胞凋亡而发挥作用。
Objective To investigate the expression of survivin protein in cardiac cancer and precancerous lesion and its relation with cell apoptosis. Methods Streptavidin-perosidase(SP) method was used to detect the expression of survivin protein in cardiac inflammation 20 cases, atypical proliferate Ⅰ 20 cases, atypical proliferate Ⅱ 16 cases, atypical proliferate Ⅲ hyperplasia 6 cases and cardiac cancer 67 cases. TUNEL was used to detect the apoptosis cells in the above-mentioned cardiac tissue, and to calculate angiogenesis index. Results ①The expression rate of survivin protein increased gradually from cardiac inflammation to the atypical proliferate Ⅰ ,proliferate Ⅱ ,proliferate Ⅲ and to cardiac cancer with deteriorating lesion, their survivin protein was ( 15.15 ± 6.97) %, ( 14.84 ± 6.66)%, ( 14.33 ±5.34) %, (9.31 ± 3.96) % and ( 6.47 ± 2.82) %, respectively, while their angiogenesis index decreased gradually 0.0%, 30.0 %, 25.0%, 83.3 % and 61.2%, respectively. ② Cardiac cancer had no outstanding difference from atypical proliferate Ⅲ in the expression of survivin protein( P〈0.05), while had outstanding difference from other cardiac tissues( P 〈0.01). There were no outstanding difference in the expression of survivin protein between cardiac inflammation and atypical proliferate Ⅰ ,proliferate Ⅱ ( P 〈0.05). ①The expression of survivin protein and apoptosis index correlated negatively( rs=-0. 912, P 〈0.05). Conclusion The gradual increase of the expression of survivin protein occurred at the early stage of carcinomatous change and played a role to inhibit cell apoptosis.
