摘要
目的:研究硝苯地平(Nif)和尼卡地平(Nic)对人胚肺成纤维细胞胶原与透明质酸(HA)合成以及N-乙酰β-氨基葡萄糖苷酶(NAG)活性的影响,为应用钙通道阻滞剂防治器官纤维化提供依据。方法:采用3H-脯氨酸掺入、放射免疫和荧光法分别测定胶原与HA合成及NAG活性。结果:Nif和Nic在10~40μmol/L浓度范围内均呈浓度依赖性地抑制胶原与HA合成;Nic在10~20μmol/L浓度时还使细胞培养上清液中NAG活性增加。结论:Nif和Nic可望成为抗器官纤维化的有效药物。
To study the effects of nifedipine(Nif)and nicardipine(Nic)on collagen and hyaluronic acid(HA)synthesis,as well as N-acetyl-β-glucosaminidase(NAG)activity of human lung fibroblasts(HLF).METHODS:The collagen and HA synthesis and NAG activity were determined by measuring the incorporation of 3H-proline,radioimmunoassay and biochemical method respectively. RESULTS:Both Nif and Nic decreased the collagen and HA synthesis in a drug concentration-dependent manner at l0~40μmol/L. Nic also increased the NAG activity in the supernatant of HLF culture at l0~20μmol/L. CONCLUSION:Nif and Nic might be a hopeful antifibrosis drug.
