放射线和TNF-α诱导胃癌细胞表达胸苷磷酸化酶的研究

摘要 目的 探讨胃癌细胞是否表达胸苷磷酸化酶(Thymidinephosphotylase,TP)以及放射线照射和肿瘤坏死因子作用后能否诱导胃癌细胞使其高表达TP。方法培养BGC823胃癌细胞株并按0Gy、1Gy、5Gy、10Gy、20Gy、30Gy的剂量梯度进行照射并培养12h后收集细胞;按30GY照射后培养1h、3h、6h、12h,24h后收集细胞;放入肿瘤坏死因子使其在培养液的终浓度达到10ng/ml并培养3h、6h、12h、24h后收集。然后分离胞浆蛋白,测定浓度后用蛋白印迹方法检测TP含量变化。结果蛋白印迹检测发现正常培养的BGC823胃癌细胞,经放射线照射和肿瘤坏死因子作用的BGC823胃痛细胞均未见目标TP显像。结论BGC823科癌细胞不仅不表达TP而且也无法用放射线和肿瘤坏死因子诱导后使其表达TP。 AIM To study whether thymidine phosphorylase(TP) is expressed and further up- regulated by gamma- ray irradiation or TNF- αin gastric cancer cell line BGC-823. METHODS Gastric cancer cell line BGC-823 was treated with 60Co gamma - ray external irradiation at doses from 0 to 30Gy, or with TNF- αat final concentraion of 10ng/mL. The cells were collected at different intervals from 0 to 24 hours after the treatment, and plasmosin TP was detected with Western Blot method. RESULTS TP was not expressed even after treatment with gamma-ray or TNF- αin gastric cancer cell line BGC- 823 plasm. CONCLUSION Gastric cancer ceil line BGC - 823 does not express thymidine phosphorylase even after induction with gamma - ray or TNF - α.
