摘要 目的探讨重组人类转化生长因子β1(rhTGF-β1)对人近端肾小管上皮细胞(HKC)桩蛋白(Pax)表达时相的影响。方法将体外培养的HKC随机分为4组。对照组(C组)无血清培养基(FSM)培养;rhTGF-β1培养组(T1、T2、T3组)分别用含10ng/mlTGF-β1的FSM培养,取不同时相点(T1组24h,T2组48h,T3组72h)终止培养。MTT法检测rhTGF-β1诱导HKC增殖的时效关系,RT-PCR检测HKC的PaxmRNA表达,免疫组化和Westernblot检测Pax蛋白的表达。结果C组可检测到PaxmRNA和蛋白表达;PaxmRNA和蛋白表达T组明显高于C组,且T2、T3组明显高于T1组(P<0.01)。结论TGF-β1诱导HKC合成Pax具有时间依赖性。 Objective To explore the effect of transforming growth factor β1 (TGF-β1) on expression of paxillin(Pax) in human kidney proximal tubular epithelial cell line (HKC cells) in different phases. Methods HKC cells cultured in vitro were divided into four groups at random. Control group(C group): HKC cells were cultured with serum free medium (FSM), and TGF-β1-treated groups (T1, T2, T3 group) : HKC cells were cultured with FSM containing 10ng/ml TGF-β1. In the latter groups, duration of treatment varied as follows: T1 group for 24h, T2 group for 48h, T3 group for 72h. The proliferation of HKC cells induced by TGF-β1 was assessed by MTT and the expression of Pax was determined. The gene expression of Pax was determined by reverse transcripton polymerase chain reaction (RT-PCR) and the protein expression of Pax was assessed by immunohistochemieal staining and Western blot. Results TGF-β1 could induce the proliferation of HKC cells in a time-dependent manner. In C group there was expression of Pax mRNA and protein in HKC cells. The expression of Pax mRNA and protein in HKC cells was positive in all the T groups, especially in T2 group and T3 group they were significantly increased compared with T1 group (P〈0. 01). Conclsuon Pax mRNA and protein were induced in HKC cells with high expression by TGF-β1.
