大鼠心肌梗死急性期缺血交界区基因表达谱特征 被引量:5

摘要 目的研究心肌梗死前后心梗交界区的基因表达谱差异,寻找保护性基因和损伤相关基因,从而为研究心肌梗死的分子机制与治疗提供依据。方法结扎大鼠冠状动脉左前降支复制心肌梗死动物模型,应用基因芯片技术研究心肌梗死24小时后心肌梗死交界区基因表达谱的变化特征。结果冠状动脉结扎所致心肌梗死,在模型/假手术梗死交界区基因表达谱的研究中共筛选出190条差异表达基因,其中下调89条,上调101条。涉及已知功能的12类基因,主要与能量代谢、信号转导、物质运输等途径相关。结论急性心肌梗死后表现出多环节的功能失调,存在多条途径对相关基因表达进行调控。 Objective To study the molecular mechanism of acute myocardial infarction (AMI), searching for protective genes and injury - related ones, aimed to fight myocardial infarction disease. Methods Rat model of AMI was established with left anterior descending (LAD) coronary artery hgation. Gene chip was used to verify the mechanism of AMI at molecular level. Results The author screened out 190 differential expression genes between AMI rat myocardiurn and sham operated rat myocardiurn in the gene expression profile study; In the Model/Sham operation gene expression profile, there were 89 genes depressed expression and 101 genes elevated expression. Most of them involve in the 12 known categories, and relate to the metoblolism, signal and trandport pathways. Conclusion The major possible mechanism of AMI is to regulate the locall expression of related gene through a multi - pathway.
