摘要
目的探讨血管内皮生长因子(VEGF)反义寡核苷酸对裸鼠骨肉瘤生长的抑制作用。方法制备BALC/C裸鼠皮下骨肉瘤模型18只,随机分为3组:VEGF反义寡核苷酸(ASODN)治疗组、VEGF正义寡核苷酸(SODN)治疗组及对照组,每组6只。接种MG-63人成骨肉瘤细胞后24 h内,分别皮下注射ASODN及SODN进行治疗,对照组只注射生理盐水,每周2次,连续4周;观察各组裸鼠肿瘤的生长情况、裸鼠的一般情况与生存期。断颈处死裸鼠,游标卡尺测量肿瘤体积大小,天平称量肿瘤的重量。结果与对照组瘤重(7.45±0.60 g)比较,VEGF ASODN组(4.13±0.80 g)能明显抑制裸鼠肿瘤生长(P<0.01),VEGF SODN组(6.92±0.69 g)则无明显抑制作用(P>0.05)。VEGF ASODN组、VEGFSODN组抑瘤率分别为44.56%、7.11%。结论VEGF反义寡核苷酸能抑制裸鼠体内骨肉瘤生长,通过反义寡核苷酸下调VEGF的表达,可达到治疗骨肉瘤的目的,为骨肉瘤的基因治疗提供了实验依据。
Objective To study the inhibitory effect of VEGF antisense oligonucleotides on the growth of osteo- sarcoma in nude mice. Methods The osteosarcoma cells were cultured and implanted subcutaneously into 18 nude mice which were subsequently divided into three groups: VEGF ASODN group, VEGF SODN group and the control group. Different treatment were given respectively at 24 hours after cell inoculation. The weight and volume of subcutaneous tumors were measured. Results In comparison with the tumor weight of the control group(7.45 ± 0.60 g), the tumor growth of VEGF ASODN group(4.13 ± 0.80 g) was significantly inhibited. The inhibition rate of tumor growth of the VEGF ASODN group and VEGF SODN group was 44.56% and 7.11%, respectively. Conclusion It was demonstrated that the growth of osteosarcoma in nude mice can be inhibited by VEGF antisense oligonucleotides. The suppression of VEGF expression by ASODN may offer both a therapeutic approach and an important theoretic foundation for gene therapy against osteosarcoma.
