摘要
目的:探讨CD134/CD134L共刺激信号在SLE发病机制中的可能作用及狼疮方对其表达的影响。方法:采用狼疮样BXSB小鼠模型,随机分为3组:狼疮方治疗组、泼尼松治疗组、空白模型组,每组6只,疗程10周。另设与BXSB小鼠同基因的正常C57BL/6小鼠6只为正常对照组。分别采集上述各组小鼠外周血和脾组织进行检测。结果:(1)与正常对照组比较,模型组BXSB小鼠的血清IgG和抗ds-DNA抗体水平和脾组织CD4+CD134+、CD8+CD134+、CD19+CD134L+、CD23+CD134L+的百分比及CD134、CD134L的mRNA拷贝数都显著增高(P<0.05或P<0.01)。(2)狼疮方治疗组或泼尼松治疗组的血清IgG、抗ds-DNA抗体水平和脾组织CD4+CD134+、CD8+CD134+、CD19+CD134L+、CD23+CD134L+的百分比及CD134、CD134L的mRNA拷贝数都显著低于模型组(P<0.05或P<0.01),而与正常对照组比较无统计学差异(P均>0.05)。结论:狼疮样BXSB小鼠存在CD134/CD134L的异常表达。狼疮方可减轻狼疮样小鼠高丙种球蛋白血症,减少体内自身抗体产生,对CD134/CD134L存在泼尼松相当的下调作用,抑制T、B细胞的异常活化,有效地治疗SLE。
Objective:To investigate the effects of lupus recipe on gene expression and protein synthesis of CD134 and CI134 L in splenic cells of lupus- prone BXSB mice. Methods:Eighteen male lupus- prone BXSB mice model were used in the experiment. The model mice were divided into three groups: un- treated model group, lupus reeipe(LR) - treated group, and prednisone treated group. Model mice were killed in 10 weeks. The control groups consisted of 6 syngenetie normal C57BL/6 male mice. Peripheral blood and spleen tissues were collected for the study. Results: ( 1 )the serum levels of total IgG and anti - dsDNA antibody in un - treated model group were higher than that in other groups. And there were no differences among LR- treated group, prednisone- treated group and control group. (2)The positive cell percentages of CD4^+CD134^+、CD8^+CD134^+、CD19^+CD134L^+、CD23^+ CD134L^+ and the CDI34, CD134 mRNA expressions from un - treated model group were significantly higher than those from control groups(P〈 0.05, P〈 0.01 ). (3)The LR or prednisone significantly reduced the positive cell percentages of CD4^+CD134^+、CD8^+CD134^+、CD19^+CD134L^+、CD23^+ CD134L^+ and the CD134,CD134L mRNA expressions in LR - treated group or prednisone - treated group( P 〈 0.05, P 〈 0.01 comparing with model group), while the difference between the treated group and the normal group was not significant. Conclusion:BXSB mice has abnormal expression of CE134 and CD134L. Lupus recipe reduces the serum levels of IgG and auto - antibody production and can treat systemic lupus erythematosus effectively.
