阿卡波糖生物合成和发酵工艺研究进展 被引量:17

摘要 阿卡波糖是治疗Ⅱ型糖尿病的一线药物。其在游动放线菌SE50中的生物合成过程是在由Acb基因群编码的一系列酶的催化作用下完成的,包括氨基环醇的生成,4-氨基-4,6-二脱氧葡萄糖的合成和之后的糖基化作用生成阿卡波糖。以游动放线菌作为阿卡波糖的产生菌时可以通过控制培养液中麦芽糖的浓度和发酵液的渗透压来提高阿卡波糖的产量。组分C是在发酵的后期由阿卡波糖直接转化而来的,给下游的分离纯化造成了困难,如在发酵液中添加微量的阿卡波糖类似物抑制剂可转化这一反应,其中Valienamine显示了最大的抑制作用。
