摘要
目的血清睾酮酶联免疫吸附分析试剂盒的研制。方法用人工合成抗原睾酮-3-（O-羧甲基）肟：BSA免疫新西兰大耳白兔，制备出睾酮抗体并测定其效价及亲和常数。在此基础上，采用竞争性结合法，研制血清睾酮酶联免疫吸附分析试剂盒，并对所研制的试剂盒进行方法学鉴定。结果睾酮抗血清滴度1：50000，Ka=1．22×10^9L／M（M表示摩尔数）。睾酮酶免试剂盒：灵敏度0．1ng／ml；精密度：批内、批问变异系数分别为CV=3．4％和CV=7．1％。确定了男女性正常值范围：女性为0．15～1．00ng／ml，平均值为0．46ng/ml；男性为1．21～11．0ng/ml，平均值为6．6ng/ml。结论本试剂盒适于临床常规检验和科研工作需要，以国产代替进口，具有广泛应用前景。
Objective To prepare the enzyme- linked immunoadsorbent assay （ELISA） kit for serum testosterone, Methotis New Zealand white rabbit was immunized by testosterone - 3 - （O - carboxymethy） oxime： BSA as human immunogens.The antisera to testosterone were prepared.On the basis, testosterone enzyme- linked immunoadsorbent assay kit was prepared. Results Thetiters of antisera for testosterone were 1：50000; Ka=1.22×10^9L/M.Sensitivity： 0.1 ng/ml; Precision： The interand intra- assay coefficients of variation were： CV = 3.4% and CV= 7.1%.The normal range was male, 1.21 - 11.0 ng/ml, -↑x = 6.6 ng/ml; female, 0.15- 1.00 ng/ml, -↑x =0.46 ng/ml. Conclusion The kit is suitable to clinical test and scientific study.
