柱层析预处理RP-HPLC法测定女贞子中红景天苷的含量 被引量:6

摘要 目的:建立女贞子中红景天苷含量测定的RP-HPLC法。方法:样品溶液经大孔树脂和聚酰胺柱层析纯化,采用Hypersil C18色谱柱(4.6mm×150mm,5μn),流动相为甲醇:水(20:80),流速0.4mL/min。检测波长220nm。结果:红景天苷在1~80gg/mL范围内线性关系良好(r=0.9999),高、中、低浓度(40、10、5μg/mL)加样回收率分别为(101.4±1.52)%、(95.6±1.45)%、(100.5±2.18)%(n=5)。浓度为80、40、10μg/mL的红景天苷日内RSD分别为0.28%、0.17%、0.48%(n=5);日间RSD分别为0.56%、0.06%、0.12%(n=5)。稳定性的RSD=1.64%,表明红景天苷在24h内稳定。应用该法测定女贞子中红景天苷的含量为0.718mg/g。结论:本法简便、快捷、准确,适用于女贞子中红景天苷的含量测定。 Objective: To establish a RP HPLC method for determining the content of salidroside in fruits of Ligustrum lueidum Ait.. Methods: The sample was purified by macroreticular resin and polyamide column. The RP HPLC separation was accomplished on Hypersil C18 column (4.6 mm×150 mm,5μm) with mobile phase of methanol : water (20:80) and flow rate of 0.4 mL/min. The detection wavelength was 220 nm. Results: The linear correlation of salidroside was observed within the range of 1-80μg/mL (r=0. 999 9). The recovery rates of high, middle and low concentrations (40, 10, 5μg/mL) in samples were (101. 4±1. 52)% , (95.6±1.45)% and (100.5±2.18)% (n=5), respectively. The intra-day relative stand- ard deviation for concentrations of 80, 40 and 10μg/mL, were 0. 28% ,0. 17% and 0. 48% (n=5), respectively; and inter-day RSD were 0.56% ,0.06% and 0.12% (n=5 ), respectively. The stability RSD was 1.64%, so salidroside was stable in 24 hours. The content of salidroside extracted was 0. 718 mg/g by the determination. Conclusion: This method is proved to be simple, rapid, accurate and suitable for the quality control of salidroside in Fructus Ligustrum Lucidum Air.
