摘要
对能够提高免疫功能的香菇多糖测定其平均分子量。用分析型超速离心机沉降法5次测定,求出香菇多糖溶液的沉降系数S,再换用扩散泵,5次测定,求出该溶液的扩散系数D。通过公式MW=RTS/[D(1-ρυ)]求出分子量。式中MW为重均分子量,R为气体常数,T为绝对温度,υ为偏微比容,ρ为溶剂密度,计算出香菇多糖的重均分子量为36000道尔顿。为新药申报中有关质量研究,质量标准制定等项目资料,提供必要的方法和数据。
Molecular Weight of Lentinus Edodes Polysaccharide (LEP) which can improve immunity function was determined. Subsidence cofficient (S) of LEP solution was gaind through 5 times works on anatysis supervelocity centrifuge. After changing rotor into Diffusion pump Diffusion coefficient (D) was floated as similar operation. Its Weight Average molecular Weight(^-Mw) was calculated with formula ^-Mw=RTS/[D( 1-ρv], (in it, R: gas constant. T:absolute temperature v:declination slight specific volume, ρ:solvent density). So ^-Mw of LEP was 36 000 Dolton, The result is very useful for new drug application about qualitative research and qualitative standard constituting and so on.
