摘要
目的:观察糖肝煎(TGJ)对2型糖尿病并脂肪肝大鼠相关指标的影响。方法:小剂量链脲佐菌素(streptozocin,STZ)注射加高脂饲料喂养,建立2型糖尿病并脂肪肝大鼠模型。分别给予糖肝煎(糖肝煎组)、文迪雅(马来酸罗格列酮片、GlaxoSmithKline公司,文迪雅组)和生理盐水(模型组),与正常大鼠(正常组)对照。测空腹血糖(FBG)、负荷后血糖(P2hBG)、空腹胰岛素(FINS)、负荷后胰岛素(P2hINS)、肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)、脂联素;以稳态模型法计算胰岛素抵抗指数(HOMA-IR);肝脏切片分别以苏木素-伊红(HE)及苏丹三醇染色,光镜下观察。结果:糖肝煎组与模型组相比,FBG、P2hBG、FINS、P2hINS、TNF-α、IL-6、脂联素、HONM-IR等指标有显著改变(P<0·05或P<0·01),与文迪雅组相比差异无显著性意义(P>0·05)。糖肝煎组肝组织结构有显著改善。结论:糖肝煎对2型糖尿病并脂肪肝模型大鼠,能有效地改善胰岛素抵抗(IR),改善肝组织病理,其机制可能与改善TNF-α、IL-6、脂联素的表达有关。
Objective: To investigate influence of the TGJ decoction on treatment of type 2 diabetes mellitus mouse with fatty liver, try to find out the mechianism of TGJ decoction through animal experiment. Methods: The model mouse of tyep 2 diabetes mellitus mouse with fatty liver was established by injecting little dose STZ and feeding high fat food. Twenty-four of thirty two mice were feeded TGJ (A group), Wen Di Ya (B group), Sodium Chloride (C group), respectively, and compared with normal mouse. FINS, Ph2INS, TNF-α, IL-6, adiponectin, HOMA-IR, pathological section of mouse liver were observed at the end of the experiment. Results, FBG, P2hBG, FINS, P2hINS, TNF-α, IL-6, adiponectin, HOMA-IR had significantly different between A group and C group (P 〈0.05 or P 〈0.01). While between Wen Di Ya group and C group had not ( P 〉0.05). The histopathology had obvious improvement in A group after treatment. Conclusion: TGJ is a good decoction for improving insulin resistance, pathological section of liver, their mechanism may be to improve the expression of their TNF-α, IL-6, adiponectin.
